ESTUDIO EN FASE I, ABIERTO Y MULTICÉNTRICO, DE HMPL-306 EN NEOPLASIAS MALIGNAS HEMATOLÓGICAS AVANZADAS CON MUTACIONES DE LA ISOCITRATO DESHIDROGENASA (IDH)
Los sujetos pueden ser inscritos en este estudio sólo si satisfacen todos los criterios siguientes (NOTA: Esta lista no es exhaustiva):
Sujetos de edad ≥18 años.
Estado de rendimiento ECOG ≤ 2
Sujetos con neoplasias hematológicas avanzadas recidivantes, refractarias o resistentes, como se define a continuación:
Sujetos con mutación IDH documentada por secuencia de próxima generación (NGS) local o institucional.
Los sujetos deben ser refractarios o intolerantes a las terapias establecidas
Subjects are not eligible for enrollment into this study if they meet any of the following criteria (NOTE: This is not an exhaustive list):
- Subjects who received an investigational agent <14 days prior to their first day of study drug administration
- Subjects who are pregnant or breastfeeding
- Subjects with an active severe infection, some treated infections and with an expected or with an unexplained fever >38.3°C during screening visits or on their first day of study drug administration.
- Subjects with some current or prior heart conditions
- Subjects taking medications that are known to prolong the QT interval may not be eligible
- Subjects with immediately life-threatening, severe complications of leukemia such as uncontrolled bleeding, pneumonia with hypoxia or shock, and/or disseminated intravascular coagulation
- Some subjects with some current or prior gastrointestinal or liver diseases
- Subjects with inadequate organ function as defined by the protocol
HMPL-306 es un inhibidor dual de IDH1/2
Se trata de un estudio de fase 1, abierto, multicéntrico y de un solo brazo para evaluar la seguridad, la tolerabilidad, la FC, la EP y la eficacia preliminar de HMPL-306 administrado por vía oral en el tratamiento de sujetos con neoplasias hematológicas avanzadas recidivantes, refractarias o resistentes que albergan mutaciones de IDH (o co-mutaciones).
El estudio consta de 2 partes: una parte de escalado de dosis (Parte 1) y una parte de ampliación de dosis (Parte 2). La parte de escalada de dosis determinará la MTD/R2PD. La parte de expansión de dosis administrará la MTD/RP2D a sujetos con neoplasias hematológicas mIDH-positivas, incluyendo, pero sin limitarse a, LMA, SMD/NMP, AITL.
